Immune cell 'survival' gene key to better myeloma treatments

Feb. 3, 2013 ? Scientists have identified the gene essential for survival of antibody-producing cells, a finding that could lead to better treatments for diseases where these cells are out of control, such as myeloma and chronic immune disorders.

The discovery that a gene called Mcl-1 is critical for keeping this vital immune cell population alive was made by researchers at Melbourne's Walter and Eliza Hall Institute. Associate Professor David Tarlinton, Dr Victor Peperzak and Dr Ingela Vikstrom from the institute's Immunology division led the research, which was published February 3 in Nature Immunology.

Antibody-producing cells, also known as plasma cells, live in the bone marrow and make antibodies that provide a person with long-term protection from viruses and bacteria, Associate Professor Tarlinton said. "Plasma cells are produced after vaccination or infection and are responsible for the immune 'memory' that can persist in humans for 70 or 80 years. In this study, we found that plasma cells critically rely on Mcl-1 for their continued survival and, without it, they die within two days," he said.

Dr Peperzak said the team was surprised to find that plasma cells used this as a 'failsafe' mechanism in controlling their survival. "One of the interesting things we found is that because plasma cells rapidly destroy Mcl-1 proteins within the cell yet depend on it for their survival, they need continuous external signals to tell them to produce more Mcl-1 protein," Dr Peperzak said. "This keeps the plasma cells under tight control, with Mcl-1 acting like a timer that constantly counts down and, if not reset, instructs the cell to die."

Plasma cells are vital to the immune response, but can be dangerous if not properly controlled, Associate Professor Tarlinton said. "As with any immune cell, plasma cells are really quite dangerous in many respects and need to be tightly controlled," he said. "When they are out of control they continue to make antibodies that can be very damaging if there are too many. This happens in conditions such as myeloma -- a cancer of plasma cells -- and various forms of autoimmunity, such as systemic lupus erythamatosus or rheumatoid arthritis, where there are excessive levels of antibodies."

Myeloma is a blood cancer that affects more than 1200 Australians each year, and is more common in people over 60. It is caused by the uncontrolled production of abnormal plasma cells in the bone marrow and the build up of damaging antibodies in the blood. Rheumatoid arthritis and lupus are autoimmune diseases in which the antibodies produced by plasma cells attack and destroy the body's own tissues.

Associate Professor Tarlinton said that his hope was that the discovery could be used to develop new treatments for these conditions. "Myeloma in particular has a very poor prognosis, and is generally considered incurable," Associate Professor Tarlinton said. "Now that we know Mcl-1 is the one essential gene needed to keep plasma cells alive, we have begun 'working backwards' to identify all the critical molecules and signals needed to switch on Mcl-1 and keep the cells alive. Our hope is that we will identify some point in the internal cell signalling pathway, or a critical external molecule, that could be blocked to stop Mcl-1 being produced by the cell. This would be an important new platform for diseases that currently have no specific or effective treatment, such as myeloma, or offer new treatment options for people who don't respond well to existing treatments for diseases such as lupus or rheumatoid arthritis."

This research was supported by the National Health and Medical Research Council of Australia, Multiple Myeloma Research Foundation, European Molecular Biology Organization and the Victorian Government.

Ahmadinejad unveils Iran's newest fighter jet

TEHRAN, Iran (AP) ? Iran unveiled on Saturday its newest combat jet, a domestically manufactured fighter-bomber that military officials claim can evade radar.

President Mahmoud Ahmadinejad said in a ceremony broadcast on state TV that building the Qaher-313, or Dominant-313, shows Iran's will to "conquer scientific peaks."

The Qaher is one of several aircraft designs rolled out by the Iranian military since 2007. Tehran has repeatedly claimed to have developed advanced military technologies in recent years but its claims cannot be independently verified because the country does not release technical details of its arsenals.

The Islamic republic launched a self-sufficiency military program in the 1980s to compensate for a Western weapons embargo that banned export of military technology and equipment to Iran. Since 1992, Iran has produced its own tanks, armored personnel carriers, missiles, torpedoes, drones and fighter planes.

"Qaher-313 is a fully indigenous aircraft designed and built by our aerospace experts. This is a radar-evading plane that can fly at low altitude, carry weapons, engage enemy aircrafts and land at short airstrips," Defense Minister Ahmad Vahidi said.

Some reports however suggest Iran's program relies on equipment supplied by major international defense contractors ? incorporating parts made abroad or reverse-engineered technologies into its domestic designs.

Still photos of the Qaher released by the official IRNA news agency and pictures on state TV show a single-seat jet. They described it as a fighter-bomber that can combat both other aircraft and ground targets.

Iran's English-language state Press TV said Qaher was similar to the American-made F/A-18, an advanced fighter capable of dogfighting as well as penetrating enemy air defenses to strike ground targets.

Physically, Press TV said, the aircraft resembles the F-5E/F Tiger II, a much older American design that Iran has had in its arsenal since it was supplied to the U.S.-allied regime of the Shah before Iran's 1979 revolution.

"Development depends on our will. If we don't have a will, no one can take us there," Ahmadinejad told the inauguration ceremony in Tehran. "Once we imported cars and assembled them here. Now, we are at a point where we can design, build and get planes in the air."

Iran unveiled what it said was its first domestically manufactured fighter jet, called Azarakhsh or Lightning, in 2007. In the same year, it claimed that Azarakhsh had reached industrial production stage.

Saeqeh, or Thunder, was a follow-up aircraft derived from Azarakhsh. Iran unveiled its first squadron of Saeqeh fighter bombers in an air show in September 2010.

(AP) ? The Dow stock market index flirted with the 14,000 line Friday, bringing reminders of the last time it hit that mark ? almost a different era, before the financial crisis rocked the world economy.

Propelled by reports on U.S. jobs and auto sales, the Dow Jones industrial average crossed the line and kept its ground through the early afternoon, after flitting back and forth throughout the morning. The other major stock indexes also rose.

"There's a newfound enthusiasm for the equity market," said Jim Russell, regional investment director at U.S. Bank Wealth Management in Minneapolis.

But market watchers were divided over what the Dow milestone ? or even what a Dow all-time high, which is quickly approaching ? really means. To some, it's an important booster to hearts and minds, making investors feel optimistic and thus more willing to bet on the market.

"The Dow touching 14,000, it matters psychologically," said Peter Cardillo, chief market economist at Rockwell Global Capital in New York. "It attracts smaller investors."

And smaller investors, until the past few weeks, had been shying away from stocks. In the past three weeks, though, billions have flowed into mutual funds targeting U.S. stocks, according to the Investment Company Institute. Before that, investors had been pulling more cash from U.S. stock funds than they'd deposited for every month since April 2011.

To others, though, the Dow 14,000 is nothing but a number on a board, a sign more of how traders feel than how the economy is faring. And even then, it's not even the best number on the board, some traders say. Professional investors usually pay more heed to the Standard & Poor's main index, which tracks 500 companies compared to the Dow 30. The Dow, however, is more familiar to the general public.

Joe Gordon, managing partner at Gordon Asset Management in North Carolina, wasn't celebrating on Friday. He thinks the gains won't last. The fact that small investors are finally piling back in the stock market, he said, is a sign that it's getting overhyped and ready to fall.

After the Dow hit its all-time record in 2007, it fell almost steadily and a year later had lost nearly 40 percent of its value a year later.

"It is good trivia to talk about on television and the radio," Gordon said, referring to the 14,000 mark. "It's meaningless to the average professional." And for workers still unemployed by the financial crisis, he said, "it really means nothing to them."

If there's dissent over what Dow 14,000 means, what's undeniable is that it's a rarefied event: The Dow has crossed 14,000 only 15 times in its history, and the last time was more than five years ago, on Oct. 17, 2007.

If the gains hold throughout the day and the Dow closes above 14,000, that would put it in territory even more uncommon. On just nine days has the Dow managed to stay above the 14,000 mark until the end of trading. Friday's gains also mean that the Dow is within striking distance of its all-time record of 14,164.53, which it reached on Oct. 9, 2007.

For the average investor, that was all back when the stock market still seemed like a party. Housing prices were starting to ebb but hadn't cratered. Jobs were abundant, with unemployment at 4.7 percent ? compared to 7.9 percent now. Lehman Brothers still existed. So did Bear Stearns, Wachovia and Washington Mutual.

In the afternoon, the Dow was up 152 points to 14,013. The Standard & Poor's 500 rose 15 to 1,513. The Nasdaq composite index was up 38 to 3,180.

Auto sales helped. Toyota, Ford, GM and Chrysler all reported double-digit gains for January.

The government jobs report that pushed stocks forward was mixed, but traders chose to focus on the positive. The U.S. said it added 157,000 jobs in January, which was in line with expectations. Unemployment inched up to 7.9 percent from 7.8 percent in December. But, encouragingly, the government also reported that hiring over the past two years has been higher than it originally thought.

The jobs number is based on a survey of employers, and the unemployment rate is based on a separate survey of households, which is why they can diverge.

In Europe, tentative and incremental signs of a recovery were enough to push up stocks in France, Britain and Germany. December unemployment in the European Union was lower than analysts had feared, inflation unexpectedly fell, and a survey raised hopes of some growth in the manufacturing sector.

But there were also reminders that the debt problem is far from solved. The Netherlands was also forced to take over one of its major banks, to try to stave off a collapse. In Greece, dock workers extended a strike over the government's spending cuts.

Among companies making big moves:

? Drugmaker Merck fell nearly 3 percent, down $1.15 to $42.10. Its fourth-quarter profit suffered because of competition from generic medicines against its blockbuster allergy drug Singulair.

? Insurance company MetLife rose 2 percent, or 75 cents, to $38.09, after saying it plans to buy the largest private pension fund administrator in Chile.

? Zoetis, an animal health business that Pfizer just spun off, made its debut on the stock market. It was up 21 percent, rising $5.47 to $31.47.

Relieving Headaches and Migraines | clubmom

In my last blog post I discussed the Vitamin HealthTreatment for keeping Jack Frost from nipping at your nose. Do you remember the key ingredients included in the Vitamin cocktail?

This week, I will expand on the healing benefits of the Vitamin treatment for another common health concern ? Headaches and Migraines. Is Henry hammering on your head?

Here are my top naturopathic insights, treatment suggestions and prevention tips for keeping ?JACK? and ?HENRY? silent, while you play with your? little ones.

Which of the following key causes might apply to you?? ?

  • Hormone fluctuations or imbalances
  • Neurotransmitter imbalance ? remember my simple neurotransmitter urine test?
  • Spinal misalignment
  • Muscle tension of the neck, shoulder, and back that impinges on your blood vessels and reduces oxygen rich blood flow to your brain
  • Rapid temperature and barometric pressure changes
  • Magnesium Deficiency (It has been touted as the #1 mineral deficiency in North America)
  • Dehydration (especially in the winter due to dry buildings)
  • Food sensitivities (sulphites, bananas, moldy foods, MSG, sugar, dairy, wheat?)
  • Environmental toxicity and chemical sensitivities

Supportive Self Care while Suffering?

  • Magnesium is the miracle work for headaches and migraine reduction, but dosage is important. The vitamin drip contains approximately 4mL of Magnesium delivered directly into your blood stream (Note: this is different than oral dosing). Studies show impressive healing results including reduced pain to complete relief,? and rapid disappearance of nausea, vomiting, and photophobia.
  • Epsom salt baths ? rich in magnesium which also penetrates into the blood stream.
  • Oral magnesium glycinate ? it really varies per person on dosing. The standard range is 300-800mg and it is wise to consult with a health professional before trying the higher end range.
  • Hydration. It is especially dry in most of our indoor environments. Drink more fluids (this doesn?t mean drink more tea. It has to be pure, natural, water).
  • Consult a health professional who can hone in on your individual triggers and culprits from the list above.
  • Apply lavender oil topically to your temples and pulse points to calm your nerves and stress response.
  • ?Eat clean?.? Avoid all possible food trigger culprits (dairy, wheat, corn, bananas, sugar, chemicals in foods?). Instead, consume simple whole foods rich in vitamins, antioxidants and minerals, such as brown rice, green?s, pears and berries.

I hope you can select? a few strategies from above to help halt the hammers.

In health,

Tara

JAPAN: Flood recovery boosts Honda in fiscal 2013

JAPAN: Flood recovery boosts Honda in fiscal 2013

By Graeme Roberts | 31 January 2013

Honda Motor said it all but tripled operating profit for the fiscal third quarter ended 31 December, 2012?to JPY131.9bn?yen. The 198% year on year rise was due to recovery from the impact of the Thai floods in 2011 and an increase in car sales, mainly in North America.

Operating income for the fiscal first nine months (1 April to 31 December) rose 242% to JPY408.8bn.?Nine-month net income was up 108% to JPY291.3bn.

Revenue rose JPY483bn to JPY2.4 trillion in the third quarter and from JPY5.5 trillion to JPY7.1 trillion for the nine months.

Global car sales were up to 841,000, from 622,000 in the third quarter and from 1.65m to 2.5m in the nine-month period.

Honda has maintained its operating income forecast for the full fiscal year (ending 31 March) at JPY520bn, factoring in a decline in automobile sales in Europe and China due to severe economic conditions which offset the positive impact of the recent depreciation of the yen.

It has cut its car unit sales expectation from 4.12m to 4.06m units for the full fiscal year.

Expected foreign exchange losses due to rapid depreciation of the yen since the end of 2012 were factored into the downward revision of net income forecast from JPY375bn to JPY370bn.

The dividend forecast is unchanged at JPY19yen, a JPY4 increase year on year. Total fiscal year cash payments are pegged at JPY76 per share, up JPY16.
